5-Amino-1MQ and metabolism claims: what the evidence actually supports

A recent TikTok video from creator Bab the Hams presents 5-Amino-1MQ as a small-molecule drug that can “fix how your body utilizes energy,” claiming benefits ranging from targeted fat loss to muscle growth and improved insulin sensitivity. The clip describes a mechanism involving inhibition of the enzyme NNMT (nicotinamide N-methyltransferase), which would raise NAD+ levels and activate SIRT1, a protein linked to longevity and metabolism.
What the research neighborhood actually covers
The core mechanism—NNMT inhibition leading to increased NAD+—is grounded in legitimate biochemistry. NNMT does consume nicotinamide, a precursor to NAD+, and studies in rodents have shown that NNMT expression is elevated in adipose tissue of obese mice. Inhibiting NNMT in these models has been associated with reduced fat mass and improved glucose handling. However, most of this work is preclinical, using animal models or cell cultures. Human clinical trials for 5-Amino-1MQ specifically are scarce, and no large-scale, peer-reviewed studies have yet confirmed its efficacy or safety in people.
The claim that 5-Amino-1MQ “forces fat cells to burn energy rather than store it” is an oversimplification. While NNMT inhibition may shift metabolic pathways in adipose tissue, the effect size in humans is unknown. Similarly, the assertion that it stimulates satellite cell proliferation for muscle growth is based on limited experimental evidence, not established clinical outcomes. The video’s mention of improving cholesterol and insulin sensitivity is plausible given the metabolic link, but again, human data are lacking.
Limits of the evidence
No approved drug uses 5-Amino-1MQ as an active ingredient, and it is not FDA-approved for any condition. The compound is sold for research purposes only. The video’s protocol and dosage section (not fully shown in the transcript) would be speculative, as no standardized human dosing exists. Additionally, the claim that it provides “energy without jitters” is anecdotal and not supported by controlled studies.
