Acetamide, is a fundamental structural motif widely utilized in medicinal chemistry and drug design. Its unique combination of chemical stability, hydrogen bonding acceptor and donor capacity, and ease of modification makes it a valuable functional group in the development of therapeutic agents. This review explores the role of acetamide group in enhancing drug-like properties, including solubility, membrane permeability, and metabolic stability. Acetamide moieties are frequently found in a variety of pharmacologically active compounds, such as enzyme inhibitors, anticancer agents, and anti-inflammatory drugs, where they contribute to crucial interactions at the biological target site. Additionally, the incorporation of acetamido structures into lead compounds has been shown to improve binding affinity and pharmacokinetic profiles. Recent advances in synthetic methodologies have further facilitated the incorporation of acetamido group in drug design. In this article we mainly focus on synthesis, biological activity and mechanism of action of acetamido containing FDA approved drugs. This article aims to highlight the versatility and continued relevance of acetamido as a key functional group in the discovery and optimization of modern pharmaceuticals.
