TogoPhosTAC is a biologic platform that utilizes engineered phosphatases to target specific proteins for phosphate removal. This approach aims to reduce pathological tau phosphorylation, a hallmark of tauopathies, and alleviate associated behavioral deficits. The platform leverages the specificity of engineered phosphatases to selectively dephosphorylate target proteins, thereby modulating protein function and promoting therapeutic outcomes. In this study, togoPhosTAC was evaluated in male PS19 tauopathy mice, a model of tauopathy characterized by hyperphosphorylation of tau protein. The results demonstrate that togoPhosTAC effectively reduces pathological tau phosphorylation and alleviates behavioral deficits in these mice. This study presents togoPhosTAC as a delivery-ready platform for targeted protein dephosphorylation, offering a potential therapeutic strategy for tauopathies.
