As a cofactor in energy metabolism, NAD+ is essential for the functioning of the electron transport chain, glycolysis, and other metabolic pathways. Beyond its role in energy production, NAD+ is also involved in DNA repair, epigenetic regulation, and the maintenance of telomere length.Recent studies have shed new light on the mechanisms underlying NAD+ biology, including the discovery of new NAD+-dependent enzymes and the identification of novel NAD+ biosynthetic pathways. However, a significant gap in the literature remains regarding the specific roles of NAD+ in various cellular contexts.This review aims to provide a comprehensive overview of the current state of knowledge on NAD+ and identify areas for future research.
